ABSTRACT
Nasopharyngeal carcinoma (NPC) in BALB/c nude mice (NM) was re-produced by hypodermic implanting human CNE-2 cell to the right-side of neck-backposi-tion. The changes of plasma malondialdehyde (MDA) and cAMP/cGMP ratio in NM bea-ring human NPC were examined. Meanwhile, we observed that tumor's size, plasma MDAand cAMP/cGMP ratio in NM bearing human NPC were affected by polysacchoridopetide(PSP). Results indicated that in comparison with the control group, NM bearing humanNPC showed a marked increase in plasma MDA(18.25?1.37 nmol/ml),cAMP/cGMP ratio(4.55?0.35), a marked decrease in plasma, cAMP (71.83?8.51 pM/ml) and cGMP(15.77?0.65 pM/ml), both high or low-concentration PSP inhibited NPC growth.Rate of inhibition was between 59.58 to 95.53 percent. PSP decreased plasma MDA,elevated cAMP, cGMP's levels and recovered cAMP/cGMP ratio. Our results suggestedthat the elevated level of MDA in NM bearing human NPC was closely associated withthe presence of NPC. PSP had antitumor effect, probably by means of decreasing freeradicals and recovering cAMP/cGMP ratio.
ABSTRACT
This experiment was made on 4-6 week-old nude mice(NM). The CNE-2 cells were inoculated subcutaneously into each side of the neckback for the original generation. Implanted fresh transplanted NPC tissue to NM's neck-back with cannula for next passage. We passed up to 8 generations and the transplantability was 100%. We observed dynamicrly the changes of NM's immune status after they have bearing the NPC as following: (1) The net weight was significantly reduced. (2) Splenic index(S. I.) and tumor weight(T. W.) increased. S. I. was highly related to T. W. (r=0.8395). (3) Splenic NK activity slightly increased in 5 days, began to decrease in 10 days and significantly decreased in 15 or more days. NK activity was negative related to T. W. and S. I. markedly (r=-0.7132, r=-0.8237). (4) Splenic NK cells had excellent responsiveness to human IFN in vitro in 5 days and still had some in 10 days, but it had not any after 15 days. (5) Natural proliferative ratio of tumor bearing NM's splenic cells was higher than that of control group, but the responsiveness to ConA was much lower. (6) Ai Ke Xing suppressed the growth of transplanted NPC by means of decreasing sera MDA enhancing host's splenic NK activity and increasing mononuclear infiltration of the transplanted NPC tissue. So we concluded that in NM whose T cells were deficient splenic NK cells were important immunocompetent cells for resisting the growth of transplanted NPC.
ABSTRACT
The effects of polysaccharidical peptides (PSP) on the functions of tumour infiltrating lymphocytes (TILs) in vitro was explored. The results showed that PSP, at the concentration of 37-1200?g/ml, promoted the proliferation of TILs in dose-related manner. Combination of PSP with interleukin-2 (IL-2) enhanced the cytotoxicity of TILs and reduced the dose of IL-2 used for activating TILs. The results suggested that PSP could augment the proliferation and cytotoxicity of TILs in vitro.

Ehrlich's ascites cancer (EAC) in BALB/C mice by ip transplantable EAC cell is reproduced by us. The change of serum malondialdehyde(MDA), plasma cellular cAMP and adenylate cyclase (AC) are examined in the early, middle and late phase of mice bearing EAC respectively in comparison with that of control group. Results indicate that serum MDA in mice bearing EAC is continuously increased with the progress of the disease in the remarked ways in comparison with the control group. This paper reports on the effect of the culture supernatant of NPC cell line (CNE-2S) (400?l/ml) on cAMP、IL-2、and IL-2 receptor in normal lymphocyts. The cAMP levels of lymphocytes from 11 cases was 13.77?4.92pm/1?10~6. With lymphocytes suspensives added to the CNE-2S, the cAMP levels was increased to 26.1? 15.14Pm/1?10~6(P0.05)These results show that the immune function abnormality of the patients with NPC relates to the fact of that the NPC immunologic inhibitory factor declines the ability of lymphocytes to produce IL-2.
